Tóm tắt
Gút là bệnh rất phổ biến trên thế giới. Một triệu chứng của
bệnh gút là tăng axit uric máu, xảy ra do rối lọan chuyển hóa axit uric. Khi
lượng axit uric trong máu cao sẽ kết tinh lại trong các khớp xương, dẫn đến bệnh
gút. Xanthine oxidase (XO) là một enzyme đóng vai trò quan trọng trong việc gây
ra bệnh gút. Trong giai đọan cuối của quá trình trao đổi chất của purine, enzym
này xúc tác cho phản ứng oxi hóa xanthine và hypoxanthine thành axit uric. Do đó
việc tìm kiếm những chất ức chế
được enzyme XO có ý nghĩa rất lớn trong việc điều trị bệnh gút. Theo y học cổ
truyền Việt Nam, cúc hoa trắng (Chrysanthemum sinense Sabine.) được dùng điều
trị các bệnh như thấp khớp và kháng viêm. Trong quá trình nghiên cứu tìm kiếm
những cây thuốc Việt Nam có khả năng ức chế XO, chúng tôi đã phát hiện cao
methanol từ cúc hoa trắng có họat tính ức chế XO với giá trị IC50 là 5,1 μg/mL.
Từ cao methanol, chúng tôi đã cô lập được 28 hợp chất, bao gồm 15 flavonoid, 7
dẫn xuất của acid caffeoylquinic, và 6 dẫn xuất đơn giản của phenol. Tất cả các
hợp chất cô lập được thử họat tính ức chế enzym XO, trong đó chỉ có 1 chất không
có hoạt tính. Nghiên cứu động học và mối quan hệ giữa hoạt tính - cấu trúc các
chất còn lại cho thấy sự có mặt của nối đôi ở C2-C3 trên vòng B của nhóm
flavonoid đóng vai trò quan trọng, trong khi đó ester methyl của acid
dicaffeoylquinic lại làm tăng hoạt tính ức chế XO của nhóm này.

Abstract
Gout is a common disease with a worldwide distribution.
Hyperuricemia, which associates with gout, results from the overproduction or
underexcretion of uric acid. High level of uric acid may lead to deposits in
joints, leading to acute gout. Xanthine oxidase (XO) is an enzyme which plays a
crucial role in the development of gout. During the last step of purine
metabolism, this enzyme catalyses the oxidation of xanthine and hypoxanthine
into uric acid. Thus, the XO inhibitor may become a candidate of a drug for
gout. The flower of Chrysanthemum sinense Sabine. (Asteraceae) has been used in
Vietnamese traditional medicine for the treatment of rheumatism and inflammatory
diseases. In the course of our screening program for XO inhibitory medicinal
plants from Vietnam, the methanolic extract of the flower of C. sinense
exhibited significant XO inhibitory activity with an IC50 value of 5.1 μg/mL.
Phytochemical analysis of the MeOH extract of C. sinense resulted in the
isolation of 28 compounds
including 15 flavonoids, 7 caffeoylquinic acid derivatives, and 6 simple
phenolic compounds. All isolated compounds was tested on XO inhibitory activity,
one of them was ineffective in inhibiting XO. The kinetic and structure–activity
relationship studies on active compounds showed that the presence of double bond
between C2-C3 in the ring B of flavonoids plays a crucial role, while the
methylation of the carboxyl group of dicaffeoylquinic acids promotes the XO
inhibitory
