Schizosaccharomyces pombe là đối tượng nghiên cứu mới trong
sinh học phân tử và tế bào nhưng các nghiên cứu chi tiết về chức năng protein
của S. pombe vẫn chưa hoàn chỉnh, trong đó có lipase. Lipase chúng tôi thu nhận
từ S. pombe được tinh sạch bằng sắc ký lọc gel Sephadex G-75, có trọng lượng
phân tử trong khoảng 51–55 kDa. Đây là loại lipase acid với pHopt 5,5 và topt
350C. Hoạt tính của lipase ổn định khi bảo quản ở nhiệt độ dưới 200C. Nguồn cơ
chất tốt nhất cho hoạt động của lipase là dầu cá hồi nhũ hóa. Các ion Ca2+, Na+
và K+ là các chất tăng hoạt trong khi Mg2+, Mn2+ và Fe3+ lại kìm hãm hoạt tính
lipase. EDTA và các chất tẩy cũng là những chất kìm hãm mạnh đến hoạt tính
lipase S. pombe. Theo tìm hiểu của chúng tôi, đây là nghiên cứu đầu tiên trong
việc khảo sát hoạt tính lipase S. pombe. Những kết quả trên là cơ sở cho việc
tạo ra lipase tái tổ hợp từ S. pombe.

Abstract
Schizosaccharomyces pombe has taken up as a model organism,
largely in the field of cell biology and molecular biology. However, S. pombe
still lacks well known of protein function, including lipase. The lipase from S.
pombe, purified by Sephadex G-75 column chromatography, had molecular weight
approximately 51–55 kDa on SDS–PAGE. S. pombe produced acidic lipase which had
the optimum activity at 350C and pH 5.5. The lipase activity was very stable at
the temperature below 200C. The highest lipase activity was measured with salmon
oil emulsion as substrate. Lipase S. pombe activity was found to be enhanced by
Ca2+, Na+ and K+ while Mg2+, Mn2+, and Fe3+ inhibited the activity. EDTA and
surfactants also strongly inhibited the enzyme activity. To the best of our
knowledge, this is the first report describing the characterization of lipase S.
pombe. These results are the early step to prospect for novel lipase using gene
cloning.
