Tóm tắt
Trong một công trình trước đây, những thí nghiệm sơ khởi về
nuôi cấy bao phấn và hạt phấn đã được mô tả. Bao phấn (chứa tiểu bào tử ở giai
đoạn đơn nhân muộn) được giữ ở 4 oC trong 2 ngày, và sau đó được nuôi cấy trên
môi trường MS có bổ sung picloram 2 mg.l- 1 and 2 mg.l-1 zeatin 2 mg.l-1. Mô sẹo
xuất hiện từ các vết nứt của bao phấn sau 4 tuần nuôi cấy trên môi trường này.
Chồi xuất hiện từ mô sẹo sau 4 tháng trên môi trường MS có bổ sung picloram 0,1
mg.l-1 and zeatin 0,01 mg.l-1. Công trình này được thực hiện nhằm giải thích vai
trò của các chất điều hòa tăng trưởng thực vật trong sự tạo mô sẹo sau xử lý với
nhiệt độ thấp, đồng thời phân tích sự tăng trưởng của các cây đơn bội có nguồn
gốc từ hạt phấn so với các cây lưỡng bội có nguồn gốc từ sự nuôi cấy chỉ nhị.
Giberelin có thể thay thế một phần cho tiền xử lý lạnh.

STUDY ON EFFECT OF COLD-PRETREATMENT IN CALLUS
INITIATION AND ANALYSIS OF GROWTH OF HAPLOID
PLANTLETS DERIVED FROM ANTHER CULTURE IN LILIUM SP.

Abstract
In a previous paper, preliminary experiments for anther and
pollen culture in Lilium sp. cv. Sorbonne were described. Anthers (containing
microspores at late-uninucleate stage) were stored at 4 oC for 2 days, and then
cultured on MS medium supplemented with 2 mg.l-1 picloram and 2 mg.l-1 zeatin.
Callus appeared within dehisced anthers after 4 weeks of culture on this medium.
Shoots were observed from the callus, after 4 months of culture on MS medium
supplemented with 0.1 mg.l-1 picloram and 0.01 mg.l-1 BA. In this paper,
attempts were made to explain roles of plant growth regulators in callus
initiation after pretreatment at low temperature, and to analyze the growth of
haploid plantlets derived from pollens in comparison with that of diploid
plantlets derived from the culture of stamen filaments. It was possible to
replace a part of cold-pretreatment by gibberellin treatment.
