Enzym
cố định thuận lợi trong việc tái sử
dụng, tự động hóa và khả năng thu hồi
lại enzym. Calcium alginate có khả năng cố
định enzym bằng phương pháp bẫy, hạt
calcium alginate hình thành hệ thống matrix là do phản
ứng giữa sodium alginate với ion Ca2+, hệ
thống này của hạt phụ thuộc vào nồng
độ alginate tham gia phản ứng. 3 enzym trong thí
nghiệm là pepsin, amylase và invertase. Trọng lượng phân
tử pepsin (3.4.23.1, hãng Merck) vào khoảng 35000Da,
α-amylase (3.2.1.1, canh trường Viện Sinh học
Nhiệt đới) là 50000 – 65000Da và invertase (3.2.1.26,
chế phẩm nấm men Cty Cát Tường) vào
khoảng 135000 – 270000Da. Mục đích thí nghiệm này là
bước đầu xác định nồng độ
alginate phù hợp trong tiến trình cố định các
enzym có trọng lượng phân tử khác nhau. Một
số kết quả trong thí nghiệm này như sau :
Nồng độ alginate 3,5% phù hợp cho việc cố
định pepsin, α-amylase và invertase. Pepsin cố
định : Hiệu suất cố định protein –
enzym là 41,24% và hiệu suất hoạt độ riêng
cố định là 35.64% tại nhiệt độ 600C.
α-amylase cố định : Hiệu suất cố
định protein – enzym là 24,15% và hiệu suất hoạt
độ riêng cố định là 31,67% tại nhiệt
độ 550C. Invertase cố định : Hiệu
suất cố định protein – enzym là 64,27% và hiệu
suất hoạt độ riêng cố định là 39,14%
tại nhiệt độ 550C. DETERMINATION PROTEIN –

Abstract Immobilization of the enzyme permits their reuse and facilitates their
handing and recovery. Calcium alginate is a suitable matrix for enzyme
immobilization by entrapment method. The matrix systems were made from calcium
alginate beads by the reaction of sodium alginate and Ca2+ ion which
affect alginate solutions in various concentrations and molecular weight of
enzyme in the entrapment method. Three enzymes in this experiment are pepsin,
amylase and invertase. Molecular weight of pepsin (3.4.23.1, Mecrk) is about
35000 Da, α-amylase (3.2.1.1, cultural
solid of Institude of Biology and Tropic Ho Chi Minh City) is about 50000 –
65000 Da and invertase (3.2.1.26, yeast
product of Cat Tuong Limited) is about 135000 – 270000 Da. The objective of the present
study was to find the optimal conditions for calcium alginate gel
immobilization of these enzymes. Some results of this study are the following : Optimal concentration of
alginate solution is 3.5% (w/w) for pepsin, amylase and invertase
immobilization. Immobilized pepsin : Protein – enzyme yield is 41.24% and
specific activity yield is 35,64% at 600C. Immobilized
α-amylase: Protein – enzyme yield is 24.15% and specific activity yield is
31.67% at 550C. Immobilized invertase : Protein – enzyme yield is
64.27% and specific activity yield is 39.14% at 550C. |
