Biến nạp di truyền
dựa trên khả năng tự nhiên của Agrobacterium tumefaciens là chuyển đoạn DNA từ Ti plasmid của nó, T-DNA, vào tế bào thực
vật và nấm và hòa
nhập ngẫu nhiên vào nhiễm
sắc thể trong nhân tế
bào. Tiến trình chuyển T-DNA phụ thuộc vào biểu hiện của gen vir (gen
gây độc) của A. tumefaciens. Sự biểu hiện của gen gây độc
này được cảm ứng bởi sự tiết ra của
một số chất từ vết thương thực vật như Acetosyringone (AS). Sự có mặt của
biên trái và biên phải
ở T-DNA làcần thiết
trong tiến trình này. Trong nghiên cứu
này, chúng tôi đã biến
nạp thành công vào nấm
bệnh cây Phytophthora palmivora thông qua sự biến nạp gián tiếp nhờ A. tumefaciens bằng phương pháp nuôi cấy chung giữa nấm và vi khuẩn A. tumefaciens mang plasmid chứa T-DNA có gen hph
kháng hygromycin B để chọn lọc nấm biến nạp. T-DNA đã hòa nhập
vào trong bộ gen của
nấm và ổn định qua các thế hệ
nuôi cấy nấm. Sự hiện diện của gen hph trong
DNA bộ gen của P. palmivora được
kiểm tra bằng PCR. Nghiên cứu này là
phương tiện
để phân tích tính gây
bệnh của nấm P. palmivora trong tương lai và thúc đẩy
sự sử dụng phương pháp biến nạp hữu dụng này rộng rãi hơn trong nghiên cứu về vi nấm.

Abstract:
Agrobacterium
tumefaciens - mediated genetic transformation is
the transfer of T-DNA of Ti plasmid in A.
tumefaciens into plant and fungal cells and the
T-DNA randomly is integrated into the chromosomes of cell nuclei. The T-DNA
transferring process depends on vir genes of A. tumefaciens.
Expression of the virulent genes is induced by secretion of some substances
from plant wounds such as Acetosyringone. The
presence of left and right borders in T-DNA is necessary in this process. In
this work, Agrobacterium tumefaciens-mediated
transformation was successfully applied to the phytopathogen
Phytophthora palmivora by coincubation method between P. palmivora and A. tumefaciens
carrying plasmids containing T-DNA with hygromycin B
resistant hph
gene to select transformants of the fungus. The T-DNA
was integrated into the genome and was stable through cell division. The
presence of hph
gene in P.palmivora
DNA genome was checked by PCR. These findings should facilitate future analysis
of P.palmivora
pathogenicity and stimulate wider use of this
valuable transformation method in fungal research.
